Oral Film Sachet Qualification: Moisture, Seal and Stability Evidence

Compare oral strip packaging through material WVTR, seal testing, finished-pack integrity and formula-specific stability evidence.

FORMULATION & SAMPLESPACKAGING DEVELOPMENTPROJECT DOCUMENTATION
Representative inspection of an oral strip aluminum sachet and its sealed edge

Qualify an oral film sachet as a finished product-and-pack system. Compare material barrier data under matching conditions, assess the sealed pack, and review stability for the actual formulation. Foil appearance or a single WVTR value leaves important questions open.

For brand, procurement and development teams. These checklists are proposed review tools; they do not report testing performed by HYTANBIO or provide personal medical advice.

Define the formula and pack that the evidence covers

Oral film sachet qualification asks whether a particular pack protects a particular formulation under its intended storage and distribution conditions. “Aluminum sachet” describes part of the construction. It leaves the laminate, sealant, opening design and finished-pack performance unspecified.

Start with the approved film revision and complete sachet construction. Record dimensions, print revision, release liner and packing configuration. If the supplier changes an adhesive, laminate source or sealant layer, ask which earlier evidence remains applicable.

Keep proposed markets and shipping routes in the brief. A room-temperature warehouse assessment does not describe every transport excursion. The project team should decide which exposures need investigation and which storage instructions the available results support.

Distinguish material barrier, seal strength and pack performance

Evidence type Question answered What it leaves unresolved
Material WVTR How water vapor passes through tested material under stated conditions Integrity of the completed sachet and its seals.
Seal-strength test How the tested joint behaves under the named method Whether the whole package has an undetected leak.
Leak / integrity assessment Whether the evaluated pack shows a defect under that test Long-term protection of each ingredient in the formula.
Packaged-product stability How the product changes in its specified pack during the study Untested formulas, revisions or storage conditions.

ASTM F1249 addresses water vapor transmission through plastic film and sheeting. ASTM F88/F88M addresses the seal strength of flexible barrier materials. Their scopes explain why a single packaging number cannot replace the qualification review. Specify the method edition with the laboratory. ASTM F1249: Water Vapor Transmission Rate Through Plastic Film and Sheeting; ASTM F88/F88M: Seal Strength of Flexible Barrier Materials.

Make WVTR comparisons on the same basis

Record temperature, relative humidity, units, specimen construction and reporting capability. A lower WVTR is a useful comparison when conditions and reporting basis are comparable. Ask whether a very low result is a quantified measurement or a value below the instrument’s reporting limit.

Use stability conditions with their original scope

ICH Q1A(R2), dated February 2003, gives a drug-stability reference of 40°C ± 2°C and 75% RH ± 5% RH for six months in its general-case accelerated framework. The guideline concerns new drug substances and products. That reference is not a universal expiry calculation or an automatic requirement for nutritional oral films. ICH Q1A(R2): Stability Testing of New Drug Substances and Products, section 2.2.7.1.

For the actual film, ask the study owner to justify conditions, pull points and tests. The panel may need to follow ingredient content as well as handling and pack condition. Define the agreed criteria before interpreting a later change.

An accelerated result can help investigate a weakness. Turning that observation into a shelf-life statement requires an appropriate evidence assessment. “Six months in a chamber equals two years on a shelf” is not a general conclusion supplied by the cited guideline.

Build a pack qualification record

  • Film formula and sachet construction revisions tied to each report.
  • Material barrier data with conditions and reporting limits.
  • Finished-seal and integrity assessments selected for the actual pack.
  • Packaged-product study plan with criteria and pull-point results.
  • Opening, liner removal and strip handling observations after storage.
  • Change-control rules for material, equipment or packing-site changes.

Record usability failures as well as analytical failures

A film may meet ingredient assay criteria but stick to its liner. Another pack may protect the film during storage yet tear unpredictably during opening. Include those observations in the sample review and define the remedy. They affect the product users receive.

Turn gaps into a defined follow-up

When comparing two sachet options, list the evidence held for each and mark the unanswered question. Request a missing test or justified explanation. A certificate for a different laminate leaves a gap in the finished-pack assessment.

Record any conditional approval. For example, artwork approval might allow a print proof to proceed while the final construction remains under review. Keeping those permissions explicit reduces the risk that a sample decision becomes an unintended production authorization.

The packaging development brief can record construction and approval stages. Read the shelf-life guide for the wider expiry decision, and use the COA checklist to connect packaging work to batch release.

Frequently asked questions

Does aluminum packaging guarantee a shelf life?

No. Review the complete sachet construction, finished seals and stability evidence for the actual formula and intended conditions.

Can two WVTR values be compared directly?

Only after checking their test conditions, units, specimen construction and reporting limits. The method and data basis need to be comparable.

Does a seal-strength result prove that a sachet has no leak?

Seal strength and package integrity are different questions. Select and justify the assessments needed for the finished package.

Does ICH accelerated testing automatically give a nutritional film a two-year expiry?

No. ICH Q1A(R2) is a drug-stability reference. A nutritional film needs a justified study and shelf-life assessment for its own product, pack and market.
